1、非洲猪瘟疫苗研究
针对世界范围内至今尚无有效预防用非洲猪瘟疫苗的严峻现状,研究建立非洲猪瘟病毒CRISPR/Cas9、Cre-Loxp基因敲除操作技术平台和病毒活载体疫苗构建平台,创制以国内流行毒株为亲本、具有良好安全性和免疫效果的非洲猪瘟病毒基因缺失活疫苗候选株和表达ASFV保护性抗原的重组腺病毒、伪狂犬病毒和猪痘病毒载体疫苗候选毒株,开展安全性、遗传稳定性和免疫效力评价试验,为非洲猪瘟疫病防控提供技术支撑。
2、非洲猪瘟新型诊断技术与免疫机制研究
研制非洲猪瘟病毒重组酶介导链替换(RAA)核酸扩增检、双抗体夹心ELISA、阻断ELISA等检测试剂盒,终建立特异、敏感、快速、简便的非洲猪瘟病毒核酸及免疫学检测技术。研究ASFV 毒力蛋白与宿主的互作、对宿主自噬、焦亡、干扰素通路、cGAS-STING等信号通路的调控,探讨其对宿主先天性免疫的影响及参与免疫逃逸的机制,为非洲猪瘟基因缺失疫苗的研发提供新思路。
3、牛结核病等人兽共患病综合防控研究
牛结核病是牛的一种危害严重的人兽共患病,对其进行“检疫+扑杀”等综合防控是控制该病的重要措施。针对不同品种的牛,集成PPD皮内变态反应实验、IFN-γ释放实验、巢式PCR方法等诊断技术,并在规模化牛场进行示范、推广与应用。筛选不同感染期的特异性蛋白,构建和表达重组蛋白,建立潜伏期和排菌期特异性标志物的诊断产品。
